Atempt to cultivate
General observations
- 05.11.2019 General observations.
On e.coli medium, nematodes are found on the cube although they are inactive, they are rarely found outside but when they are found they are really active
On nematode medium, nematodes are found in really high density outside of the inoculum cube but they are inactive. Relatively few of them are found on the cube but they remain active.
Look at the sleeping cycle of nematodes maybe hibernation.
Plate #1
- 30.10.2019 LB medium cooked by Maxi
- 01.11.2019 e.coli inoculated by Maxi
- 05.11.2019 nematode inoculum by Elliy from the original carolina plate. e.coli doesn't look grown, nematodes remain on the inoculated cube, many of them can be found.
Plate #2
- 04.11.19 e.coli medium by Elliy
- 05.11.19 inoculated by Elliy with a cube of nematodes from the carolina plate
Plate #3
- 04.11.19 e.coli medium; nematode inoculum Miga
- 05.11.19 One nematode on inoculated cube not moving. There is one other nematode on the e.coli medium, it is moving manically.
Plate #4
- 04.11.19 e.coli medium; nematode inoculum Maggy
- 05.11.19 Nematodes don’t like the e.coli medium. They all stayed on the cube most of them are dead but some are still alive. No nematodes founds outside of the inoculated cube.
Plate #5
- 04.11.2019 nematode medium. e.coli plate.
- 05.11.2019 inoculum: 1 active nematode. Outside inoculum loads of nematodes inactive. Not sterile plate.
Plate #6
- 04.11.2019 nematode medium Elliy inoculum
- 05.11.2019 Inoculum cube has a couple of active approx five nematodes, outside there is no e.coli growing, the coli wasn’t properly inoculated. No nematodes.
Plate #7
- 04.11.2019 nematode medium Maggy nematode inoculum
- 05.11.2019 Bena. One active nematode on inoculated cube, on the e.coli plate there is many nematodes in high density that are inactive.
Plate #8
- 24.10.2019 Max+miga. We find double the nutrition density.
- 05.11.2019 On the inoculum we found 20 inactive nematodes and on the e.coli nothing.
Plate #9
- 24.19.2019 e.coli medium
- 05.11.2019 A couple of inactive remains on the inoculum but not elsewhere. hypothesis< the nematodes have created a mausoleum to die peacefully without being watched by us the voyeurs.
Plate #10
- 30.10.2019 e.coli medium prepared by maxi.
- 05.11.2019 No e.coli plane, diverse bacteria colonies. The inoculum had some active nematodes. The lid taken off for inspection which caused damage in all moving nematodes. Only remains are visible.
Plate #11
Atempt to safe for future
C. elegans on plates #1-#11 kept in a fridge for a couple of weeks. Medium doesn't dry out and is kept fresh. Looks pretty much promising
C. elegans on plates #15-#17 kept in a fridge for a day and then in a freezer with minus 20C for 1 week. Plate with lb medium has an old smell. discarded. Other plates have perhaps living nematodes, continue being kept in a fridge.
